2 Method of operation
2.1 Sample Handling: Take 25 mL (g) of the sample into a sampling tank or homogenized cup containing 225 mL of sterilized phosphate buffer dilution (or saline) to make a 1:10 sample homogenate, and use 1 mL to destroy Pipette 1 mL of sample 1:1, inject into a test tube containing 9 mL of the diluted solution, shake it to become a 1:100 sample, and so on, each time change a straw.
2.2 Inoculation: 2~3 suitable dilutions are selected for general food selection. Liquid samples with low bacterial content (such as drinking pure water and mineral water) can be directly taken up for testing. Place the coliform rapid test piece on a flat test bench, and use a sterile pipette to draw 1 mL of the sample. The sample is evenly dropped onto the paper. Allow to stand for about 1-2 minutes to completely wet the paper. Inoculate two pieces for each dilution. . At the same time, make a blank negative control. 2.3 Cultivation: Place the test pieces horizontally in a constant temperature incubator, and the number of stacked pieces should not exceed 10 pieces. The culture temperature was 36 ° C ± 1 ° C, and the culture was carried out for 15 to 24 hours.

3 counting principles and reporting methods:
3.1 After the culture, the blue colonies on the paper are colonies of coliforms, and the number of colonies between 15 CFU and 150 CFU is counted.
3.2 If the number of colonies in both dilutions is between 15 CFU and 150 CFU, the average number of colonies multiplied by the dilution factor is reported as the number of coliform colonies per ml (or per gram) of the sample;
3.3 If the number of colonies on all dilution test pieces is less than 15 CFU, the average number of colonies on the test piece with the lowest dilution is multiplied by the dilution factor;
3.4 If all dilutions of the test piece are grown aseptically, multiply by less than 1 by the lowest dilution factor;
3.5 If the number of colonies on the highest dilution test piece is greater than 150 CFU, count the average number of colonies on the highest dilution test piece multiplied by the dilution factor. The reporting unit is expressed in CFU/mL (or CFU/g).

4 Additional instructions
4.1 The rapid detection of coliforms not only greatly shortens the detection time, but also the detection results are close to the traditional method, and the coincidence rate can reach about 90%.
4.2 The pH of the sample homogenate should be between 6.5 and 7.5. If necessary, adjust with 1 mol/L sodium hydroxide (NaOH) or 1 mol/L hydrochloric acid (HCl).
4.3 Note that the used test strips contain live bacteria and should be disposed of in accordance with the principles of biosafety waste disposal.

6 Storage conditions
