Intended Application

The test strip is used for detecting Cloramphenicol in honey, tissue, liver and etc.

Principle

The kit is a competitive colloidal gold immunoassay method. Sample is added to sample hole, if there is Cloramphenicol in samples, it will combine with marked antibodies, prevent the marked antibodies from combining the Cloramphenicol antigens of NC nitrocellulose membrane.

If Cloramphenicol contents in sample solution are greater than detection limit, the test line will not display color reaction and the result is positive; If Tetracycline contents in sample solution are less than detection limit, the test line will display purple and the result is negative.

Materials required But Not Supplied

l Equipments: mixer or stomacher, nitrogen-drying device, oscillator, centrifuge, measuring pipets, and balance with a reciprocal sensibility of 0.01g

l Micropipettes: single-channel 20 to 200µl and 100 to1000µl

l Reagents: ethyl acetate, N-hexane, NaOH, thick HCl, K2HPO4•3H2O.

Sample Pre-treatment

Ø Instructions

Lab gown must be clean and use disposable pipette to avoid contamination that interfere results.

Ø Solution preparation before sample pre-treatment

Liquor 1: 0.1 M K2HPO4 solution

Dissolve 11.4 g K2HPO4∙3H2O into the deionized water to 500 ml

Liquor 2: 1 M HCl solution

Take 8.6 ml thick HCl into the deionized water, thoroughly dissolve to 100ml

Liquor 3: 1M NaOH solution

Dissolve 4 g NaOH into the deionized water to 100 ml.

Ø Sample pretreatment step:

1. Honey,

u Weight 2.0±0.05g crushed homogeneous samples put into a centrifuge tube, add 4ml deionized water, 0.5ml 1 M HCl (Liquor 2) and 100 mL derivative reagent, shake well for 5min.

u Incubate in water bath at 37℃ overnight (approx. 16h) or Incubate in water bath at 50℃ (over 50℃ will affect the layering) for 3h.

u Respectively add 5ml 0.1 M K2HPO4 solution (Liquor 1), 0.4ml 1M NaOH solution (Liquor 3) and 5ml ethyl acetate, oscillate for 5min fully.

u Centrifuge at room temperature at 4000 r/min for 10 min

u Take 2.5ml supernatant into another centrifuge tube, blow dry at 50 to 60℃ with nitrogen or air.

u Add 1mlN-hexane to dissolve the dried residues, and then add 0.5ml Sample dissolving solution, oscillate for 5min fully.

u Centrifuge at room temperature at 4000 r/min for 10 min

u Take out the upper N-hexane; take 50µl supernatant for the analysis.

Concentration times: 2 detection Limits: 0.25ppb

Test Methods

l Tear test card foil pouch, remove the test card, put on a flat, clean work surface

l Use pipette to withdraw the ready sample, slowly add 3drops wisely (about 60ul) sample into the sample hole.

l Please determine the result within 3-5min. other time as invalid result.

Negative: control line(C) and test line (T) are both seen purple red

Positive: only control line(C) is seen purple red;

Invalid: control line (C) isn’t seen purple red.

Notice

² Don’t use expired or damaged products.

² The test card removed from refrigerator should place at room temperature and should be used as soon as possible to avoid failure when wet.

² Don’t touch sample hole and white nitrocellulose (NC) membrane.

² Don’t mix micropipette to avoid cross contamination

² Samples should be clear, not turbid particles and not clogged; if not, it will easily lead to blocking and inconspicuous color, which will affect the judgment of the experimental results.
