1. Principle and Application

This product is made using the principle of competitive inhibition colloidal gold immunochromatography and is used to detect tetracyclines (TCs) in tissues.

After the sample solution is dropped into the sampling well of the detection card, the tetracycline drugs in the sample solution bind to the gold labeled antibody, thereby preventing the gold labeled antibody from binding to the tetracycline drug conjugate on the cellulose membrane. When the content of tetracycline drugs in the sample solution exceeds the detection limit, the detection line shows a lighter color than the quality control line or the detection line does not show a color, and the result is positive; When the content of tetracycline drugs in the sample solution is less than the detection limit, the color of the detection line is darker or similar to that of the quality control line, and the result is negative.

4. Sample pretreatment

4.1 Instructions before sample processing:

The experimental equipment must be clean and use a disposable suction head to avoid contamination and interference with the experimental results (tissue samples such as fish, shrimp, and crabs, peeled and fat removed, homogenized with a homogenizer. Pork tissue samples, including lean meat, liver, lungs, and kidneys, should be immediately tested or collected in plastic bags for testing. If not detected in a timely manner, the samples can be stored for 24 hours at 2-8 ℃ refrigeration; it is recommended to use unfrozen tissue samples for pre-treatment experiments).

4.2 Sample preprocessing steps:

Homogenize the tissue sample (minced meat or meat puree), weigh 5g of the tissue sample, load the sample to be tested into a 15ml centrifuge tube, then add 200ul of pure water/tap water (about 7 drops of dropper) to the surface of the tissue sample, tighten the tube cap, and place the centrifuge tube vertically or obliquely (with the tube mouth facing upwards) in a 90 ℃ water bath with boiling water for 10 minutes to extract the liquid. Remove the centrifuge tube, vigorously vortex and oscillate for 3 minutes, and let it stand for 10 minutes, After the sample is cooled in a 15ml centrifuge tube, pipette 1ml of the exudate from the centrifuge tube into a 1.5ml centrifuge tube, centrifuge at 4000rpm for 3 minutes, and this supernatant is the test solution.

5. Experimental steps

5.1 Tear open the aluminum foil packaging bag of the inspection card, remove the inspection card and gold label micropores, and place them on a flat and clean table surface.

5.2 Suck the prepared sample liquid with the matching pipette, slowly and drop by drop (foam generation should be avoided) 5 drops (about 120ul) into the gold standard micropore, gently blow with the matching pipette for 30 seconds, completely dissolve the red substance, stand still and wait for reaction for 2 minutes, suck all the solution in the gold standard micropore, drop into the sample hole (S), and start timing.

5.3 Leave at room temperature for 5-8 minutes to determine the results.

6. Result judgment

Negative positive invalid

Negative: In the detection window, both the detection line (T) and the control line (C) show a purple red line, with T showing a darker color than C or T showing a similar color to C.

Positive: In the detection window, the color of the T line is lighter than that of the C line, or only the control line (C) shows a purple red line.

Invalid: Within the detection window, there is no purple red line on the control line (C).

7. Precautions

7.1 Expired or damaged aluminum foil bags are not allowed to be used.

7.2 When removing the detection card from the refrigerator, it should be restored to room temperature before opening. The opened detection card should be used as soon as possible to avoid failure due to moisture.

7.3 Do not touch the white film surface in the center of the detection card.

7.4 Droppers should not be mixed to avoid cross contamination.

7.5 The sample solution to be tested should be clear, free from turbid particles, and bacterial contamination, otherwise it may lead to abnormal phenomena such as blockage and unclear color development, which may affect the judgment of experimental results.

8. Storage and shelf life

Storage conditions: The reagent kit should be stored in a dry environment at 2-30 ℃.

Shelf life: The product has a validity period of 1 year, and the production date can be found in the packaging box.
