【Intended Application】
The test is used for detecting Avian Influenza virus antibody in Avian serum, can be used to Avian Influenza virus vaccine immunization effect assessment or auxiliary diagnosis.
【Principle】
The kit is using colloidal gold immunoassay method, sample add to sample hole, sample with colloidal gold markers will move along nitrocellulose filter membrane markers.
If the sample exist Avian Influenza virus Antibody, Antibody with colloidal gold marker will compete AIV antigen and not display purple lines, if the sample does not exist Avian Influenza virus antibody ,will display color reaction .

Store at Cool and Dry place(2-30℃)and the shelf life is 24 months.
【Sample Preparation 】
- According to the conventional method to extract 2-3 ml blood add to clean dry test tubes, coagulation at room temperature for 1 hour, centrifuge at the speed of 4000 rpm for 10-min, Separation of serum. Serum should clear, no hemolysis, no pollution. Serum sample can be kept in 2-8 ℃ short term, long term -20℃.
- Also can be used with anticoagulant whole blood for test samples. (Whole blood samples is not frozen)
【Test Methods】
- Tear test card foil pouch, remove the test card, put on a flat, clean work surface
- Use straw draw sample, add 1 drop (25ul) sample into the sample hole.
- After 10min, add 2 drop (60ul) Buffer into the sample hole.
- Read the result in 10-15 min at room temperature, more than 30 minutes the results is invalid
- (Attention: add excessive sample or buffer will appear an error result)
【Results Judgment】
- The detection threshold of this reagent is HI test 1:32. Through dilute the detected serum, find its maximum dilution which shows positive, last can get the antibody titers.
- For example: dilute the detected serum with physiological saline to do respectively 1:2, 1:4, 1:8's dilution, then detect these three different dilutions of samples, if 1:2, 1:4 were positive, while 1:8 was negative, then the maximum dilution for the positive is 1:4 , so the final antibody titers of this sample is 25 (1:32)× 22 (1:4)= 27 (1:128)
【Results Interpretation】
- For immune samples, antibody levels will reflect the immune effect of high and low
- For samples without vaccination, positive results show that there may be AIV infection, should be combined with clinical analysis and other methods.
【Test Method Limitation】
The test is used only for detecting AIV general antibody levels but for subtype analysis
【Notice】
- Please read the instructions carefully before testing, a variety of reagents only for this experiment
- Don’t use expired or damaged products.
- The test card removed from refrigerator should be return to the room temperature and should be used as soon as possible to avoid failure when wet
- Do not use tap water, normal saline and distilled water as a negative control
- Sample should be fresh and clear, avoid using turbidity, polluted hemolysis, blood lipids, viscous samples
- Avoid touch the sample hole and the membrane chromatography of window
- The waste shall be regarded as pollutants, please dispose of them in time
